Sweet! I now have enough posts to start putting hyperlinks in my messages!
My absolute favorite webcomic is Something*Positive by Randy Milholland. The humor is dry and self-deprecating. The artwork starts out rough but gets better quickly, though never reaching the level of comics like AppleGeeks. The First Comic does a good job setting the tone for the whole strip. At least take a look at that one.
My second favorite changes frequently, but take a look at The Perry Bible Fellowship. It is just sick and twisted, and wrong on so many levels.
And third, this forum already has a few vocal XKCD fans, but I'll post a link anyway. It's pretty nerdy, but I think a lot of us can really appreciate some of Randall Munroe's humor.

Oi! If we're adding non-comics to this list, check out the Despair.com demotivational posters.
This site is my favorite anti-depressant. No matter how rotten the day has been, I can always look here and feel better about life. I'm pretty sure that is messed up, but I like it.
Pennyarcade.com , Pvponline.com , Sinfest.net , Reallifecomics.com, Betterdays .
Wowwww, all of those are so mean and awesome. I love the demotivation oneOriginally Posted by Gabriel

I vote SinFest as the most quotable comic. My favorite is: "If God sends you a helpess scapegoat it's a sin not to use him."Originally Posted by Velvet-Tongue

I can't believe no one's said Achewood.
Something about God doing puppet theatre, even making fun of goth, just makes me smile.

I like Dork Tower. It's written by a British gamer, living in America. The Goths in it are all into playing Vampire the Masquerade, and there have been 2 spoof "ClanBooks", One called Clanbook: Perky and the other called Clanbook: Mopey.
Dork Tower
He's also the man behind My Little Cthulhu
Nobody's mentioned flem comics yet! Home of the Jay series. It's actually done by the guy that illustrates Two Lumps (I saw someone had mentioned that one). It's super good.
